    目的 研究马铃薯中α-茄碱的液相色谱串联质谱联用的检测方法。方法 样品经酸性水与有机溶剂的混合溶液加速溶剂萃取,中等极性阳离子交换固相萃取小柱净化,氮气吹扫吹至干后,含0.1%甲酸的乙腈+水(80+20, v/v)溶解残渣,混匀并过滤后,亲水作用的液相色谱柱进行分离,多反应离子监测定性和定量分析。对前处理溶液、液相分离条件、质谱检测基质效应进行了考察。 结果 方法检出限为0.5μg/kg,定量限为1.6μg/kg,标准曲线的线性范围为:0.1-100μg/L,精密度为3.41%-7.56%,样品加标回收率范围为82.8%-99.0%。结论 加速溶剂萃取提取简便、效率高,亲水作用液相色谱分离后,串联质谱检测灵敏且定性准确,满足风险监测需求和中毒分析要求。

    Abstract:

    Objective To establish a method for determination alpha-solanine in potatoes by liquid chromatography-tamdem quadrupole mass spectrometry. Methods The potato samples were extracted using the mixed solution of acidity water and organic solvent by accelerated solvent extraction . After the purification by MCX solid phase extraction, the eluent was dried-up with nitrogen. The analytes were dissolved by the mobile phase and separated by hydrophilic interaction chromatography and determined using multi-reaction monitoring mode. The conditions of the pretreatment solution and liquid chromatography and matrix effect were optioned Results The limits of quantification and detection were 0.5 μg/kg and 1.6 μg/kg. The linear range of standard curve were from 0.1 μg/L to 100 μg/L. The relative standard deviations of detection were at 3.41 %~7.56%, the spiked sample recoveries of analytes were at 82.8 %~99.0%. Conclusion The simplicity, high efficiency of accelerated solvent extraction and the separation effect of hydrophilic interaction chromatography and the sensitivity, accuracy of tamdem quadrupole mass spectrometry can meet the needs of risk monitoring and poisoning analysis.
